If the dark spot is just behind the curve of her belly and behind her second fin (called the anal fin) then this would be her "gravid spot". All female guppies have this spot. It's one of the ways you can tell the male guppies from the females.
This spot will get darker when the female is pregnant and as her pregnancy progresses. It may also change in size and shape. Depending on the color of the guppy it can also be different colors. It is sometimes hard to see on dark guppies, like half-blacks or blues, and may be pinkish or red in blonde guppies. If your female guppies have a very dark spot and a very fat look to them, or a spot that has been getting darker, it probably means they are pregnant. This can be the case even if you have never had a male guppy in your aquarium as female guppies can carry sperm from a male for up to 6 months.
